LAB 11 OBJECTIVES

 

1)    Identification of unknown bacteria (Period 4).

a)    Obtain all the cultures you prepared from period 3.  Use these cultures to perform the following tests.  Record all results on your Descriptive Chart.

i)             Sugar fermentation tests (phenol red glucose broth, phenol red lactose broth, phenol red sucrose broth, & phenol red mannitol broth)

ii)           Nitrate reduction test (nitrate broth)

iii)         Urea hydrolysis test (urea broth)

iv)         Phenylalanine deaminase test (phenylalanine agar)

v)           Hydrogen sulfide test (Kligler iron agar)

vi)         Oxidase test (tryptic soy agar)

vii)       Catalase test (tryptic soy agar)

viii)     Starch hydrolysis test (starch agar)

ix)         Casein hydrolysis test (skim milk agar)

x)           Lipase test (spirit blue agar)

 

2)    Identification of unknown bacteria (Period 5).

a)    Using all the data recorded on your Descriptive Chart for your unknown, determine the identity of this bacterium by using the following resources:

-         Lab manual (chapter 41 separation outlines)

-         Bergey’s Manual of Determinative Bacteriology

-         Probabilistic Identification of Bacteria Computer Program

b)    Submit your final analysis to your instructor.
